The run line is used to even out a contest by giving a numerical handicap to the favorite while giving an equal advantage to the underdog.
The run line in baseball works as follows:

As the Blue Jays are favorites in this example, they have had 1.5 runs deducted from their total score before the start of the game by the sportsbook. Meanwhile, the Orioles have been given an extra 1.5 runs to start off with. If you were to bet on Blue Jays in this case, your bet would only win if they were to win by a margin of two runs or greater. On the other hand, if you were to bet on the Orioles, your bet could still win in the event of them losing, so long as they stay within a run of the Blue Jays.
As the favorite is given a handicap, the odds offered on the run line are therefore longer than the Moneyline, which can make betting on the favorite more valuable. If you bet on the underdog on the run line, the odds are shorter than the Moneyline, but the probability of your bet winning is higher.
The over/under or total betting line is the most common example of a prop, and it involves betting on the total combined runs from both teams. If you bet on the over/under, the outcome of the game (who wins or loses) does not affect your wager at all.
With an over/under line, you will be presented with a total figure by the bookmaker. You can then either bet on the final total combined runs from both teams in the game either being over or under that among.
For example, if the over/under line is 8.5 and you bet the over, your bet will only pay out if a total of 9 runs or more are scored in the game. If you bet the under, then your bet will payout if 8 runs or less are scored come the end of the game.

Generally speaking, futures markets are released 12 months before the outcome will be decided, and can be bet on every day until then.
